Neuromedin U receptors (provisional nomenclature as recommended by NC-IUPHAR [30]) are activated the endogenous 25 amino acid peptide neuromedin (neuromedin U-25, NmU-25), a originally isolated from pig spinal cord [92]. In humans, NmU-25 appears to be sole product of precursor gene (NMU, P48645) showing broad tissue distribution, but which is expressed at highest levels in upper gastrointestinal tract, CNS, bone marrow and fetal liver. Much shorter versions NmU found some species, not human, derived least instances proteolytic cleavage longer NmU. Despite species differences structure, C-terminal region (particularly pentapeptide) highly conserved contains biological activity. S S-33) has also been identified an agonist [97]. NmS-33 is, its name suggests, 33 amino-acid protein single amidated heptapeptide identical activate NMU with equivalent potency NmU-25.
